Abstract

The present invention relates to an extraction process of silkworm gene engineering phytase with high enzyme activity. During the production of phytase in recombinant BmNPV vector-silkworm host expression system, phytase gene is used to replace polyhedron protein gene in silkworm nuclear polyhedron virus genome and special recombinant silkworm nuclear polyhedron is used to infect silkworm to produce in silkworm body target phytase protein; target disease silkworm is frozen, homogenated and absorbed with fine bran, and through drying, crushing, sieving and enzyme, activity calibration, commercial product is obtained. The process of the present invention makes it possible for the phytase in silkworm cell to release fully to result in raised enzyme activity.
本发明涉及一种高酶活家蚕基因工程植酸酶的提取方法,应用重组BmNPV载体-家蚕宿主表达系统生产植酸酶,用植酸酶基因替换家蚕核型多角体病毒基因组中的多角体蛋白基因,将特定的重组家蚕核型多角体感染家蚕,在蚕体内繁殖产生目标物植酸酶蛋白;将目标病蚕经匀浆后与一定细度的麸皮吸附,再经烘干——粉碎——过筛后标定酶活的工艺得到商业化产品,在目标病蚕匀浆处理前将病蚕作冻存处理。采用本发明的方法可使蚕体细胞内的植酸酶充分释放,较常规方法的平均检测酶活值提高55.9%以上。
