Abstract

The present invention relates to an ultrasonic auxiliary exogenous gene introduction method for soybean shoot tip conversion. In the course of using agrobacterium mediated BT gene to make soybean shoot tip conversion it adopts ultrasonic auxiliary method, and adopts the following steps: making soybean seed undergo the processes of germination and preculture, then making agroinfection, at the sametime making auxiliary ultrasonic infection treatment, co-cultivation, advertitious bud culture and induced rooting of transformed bud so as to obtain the seed produced by transformed plant. Said invention adopts the ultrasonic auxiliary method, and can raised the agroinfection efficiency, and effectively raise budding number and quantity of transformed seedlings.
一种大豆茎尖转化超声波辅助的外源基因导入方法,在农杆菌介导BT基因对大豆茎尖转化的过程中,采用了超声波辅助方法,大豆种子经萌发、预培养之后,进行农杆菌感染时辅以超声波侵染处理,再经共培养、不定芽培养及转化芽诱导生根,得到转化植株产生的种子。本发明采用超声波在外植体上制造微小伤口,增加了农杆菌与外植体之间的接触面积,在导入BT基因的过程中起到了很好的效果,明显提高了农杆菌的感染效率,有效地提高了出芽数量和转化苗的数量。
